in ned for the authentication domain username and password.
I tried domain\user but this does not work.

Are you trying to set this up under Setup->Core and Probes? If so, this is a string that is passed to the proxy server and you should be able to use the same credentials that you use when accessing the internet normally.
Have you also tried any other ways of entering a domain username and password like user@domain.com or domain.com\user or just the user? If none of those work, it would also help to know what kind of authentication the proxy server uses i.e. Basic, NTLM, Digest, OAuth etc.
