we're running PRTG network monitor but the OS is running windows 2012R2. We we do an in place upgrade to 2019 or 2022?

The windows in-place update should be transparent to PRTG, it should not affect, but we do recommend to take a backup of PRTG just in case.
You can perform a backup of the PRTG by following this guide Also, the PRTG does an auto-backup of the whole config every day. The whole configuration is stored in the PRTG Configuration.dat file and the PRTG does a daily backup of this file and saves it in the Configuration Auto-Backup folder. You can restore this configuration from previous days by following this guide
