I have Google Drive installed on a PC that is being monitored by PRTG WMI Free Disk Space (Multi Disk) and this seems to be breaking the sensor with their new virtual drive.

I tried to re-add the sensor hoping to only select the disks I want (and to ignore G) however there doesn't seem to be a way to do that (all or just 1 disk). Is there a way to exclude one drive from reporting back to the sensor?

Unfortunately, it's not possible to exclude single partitions from the WMI Free Disk Space (Multi Disk) Sensor and as the mounted partition works differently not respond to the WMI query in a proper fashion.
If you want to monitor just specific partitions, the SNMP Disk Free Sensor might be a valuable alternative.
