Are there any additional costs from a licensing perspective if we want to run your software on an EC2 instance at AWS and is this something that is supported?

In case you're not referring to our cloud offering, there are no additional costs as far as we are concerned. When you buy an on premise license it does not to us from a licensing perspective whether you run it somewhere inside your network, in AWS, Azure etc. The cost for AWS are of course yours to pay :)
Besides that, yes, it works, other customers do it as well, we basically do the same thing with our cloud offering. I order to monitor your local network we recommend to deploy a remote probe on site, because everything else would mean opening way too many ports in order to monitor the local devices from the cloud hosted PRTG server.
