is it possible to setup monitoring on users home directories and to compare directories from the prior day and if growth is more than 10%?

I'm afraid that the folder sensor of PRTG cannot compare current values with any historic measurements, sorry. You can write your own custom script to monitor the values outside of PRTG and compare the results.
