Am I able to monitor the throughput on my link using PRTG? I know PRTG can monitor traffic/bandwidth however, I will prefer if prtg can request for upload and or download a fixed sized file on a server at specified intervals. for example, if I download/upload a file of 100MBytes in 120sec that amounts to a throughput of about 6.6Mbps. Can PRTG monitor throughput? Kindly advice.

If you want to test the available bandwidth, it will be possible by creating short load peaks using the HTTP sensors of PRTG and measuring the load time. Please have a look here for more details.
Note that these sensors will utilize the available bandwidth for a short time, so choose a reasonable high scanning interval.
